Pulling the diff should be no problem for you. The cold is a differnt
problem. I've never experinced frozen grease as Jason Pritcher suggested.
It would be worth some thought. I have seen 500 HP conveyor belt motors
trip the overloads because the gear lube in the gear boxes freeze in
subzero weather.

If this were my Rover I would: (Assuming a Series II,IIa, or III)

Remove the drain plug and check how much lube, if any comes out. Should
hold 1.75 liters or 3 imperial pints or 3.5 US pints. I've had half this
amount after long trips with no ill effect.
If no lube comes out at all and its not cold enough to freeze it, its time
to pull the diff. If you get a reasonable amount of lube, I would allow as
much to drain as possible and then refill the diff. Yes it might have to be
drained again but that beats finding out you forgot to refill it later.

If this all looked acceptable I would remove the rear propshaft at the diff
and check the pinion nut as Tevor Easton mentioned. There is a torque spec
for this nut but I don't remember it.

If everything looked good or was adjusted to make it good I personally
would put it all back together and drive it.

If you want to try and narrow it down to the rear diff you could pull both
axle halfshafts and the rear propshaft. Take it for a short easy ride in 4
wheel high. I've driven mine for a couple days on just the front drive
shaft with nothing bad happening.

Bottom line, if it doesn't have lube its time to pull it for an inspection.
Two drive flange gaskets and a differential gasket, And two felt seals for
the halfshafts. Not much to replace for the peace of mind.
